The topic of the EFB lesson was ' to prepare your own antibody':
One of the two suggestions was to find a natural source for these antibodies.
The first step is to force cells to produce antibodies. This can be made by injecting antigens into an organism e.g. Mouse:
B-Cells produce antibodies.
Isolate a single B-cell and cultivate the B-cell
And finally to purify these antibodies.
Second possibility: Buy the antibodies.
But how to prepare an antigen?
To solve this problem our class has developed a scheme:
- Isolate the cDNA coding for hCG by PCR
- Clone it into an expression vector
- Produce competent cells (e.g. E.coli or S.cerevisiae)
- Transform the cells
- Induce expression of hCG
- Purify the protein
